You can modify /etc/mysql/my.cnf and dpkg/apt-get will not overwrite it.
Although, of course, it asks for it when I edit the master copy in my packages.
Maybe we switch to "ucf" somewhen which would "intelligently" insert new 
options and preserve the rest of the config file.

If you remove the line it listens on all interfaces. Feel free to
open a bug report on http://bugs.mysql.com, maybe the implement
specifiying more than one interface somewhen.
